
In the distant future, people build their homes and raise domestic animals in subterranean caverns. As a result, they live in fear of the occasional earthquake and cave-in, and have been for hundreds of years. Jeeha is one such village. One day, a boy named Simon happens to find a small, shiny drill whilst digging to expand the village. At the same time, Kamina, a guy who believes that there is another land above the village, persuades a gangster to break through the ceiling in order to leave the cavern. Suddenly, an earthquake occurs and the ceiling collapses, followed by a big robot breaking through the ceiling. Kamina is now sure that there is a world above them. Recklessly, he begins to fight against the robot. At that moment, another person appears from the land above. It is a girl by the name of Yoko. With a rifle in hand, she had been tracking the robot. However, all she could do...

Kamina is jailed for trying to escape to the surface. Suddenly a mysterious women named Yoko comes from aboves to the underground village while fighting a giant robot. Simon helps fend off the robot with the help of his new "Gunmen" which he found while drilling around.

Now above ground Kamina Simon and Yoko find themselves faced with 2 more gunmen. They dispatch these 1 of them with the help of people from Yokos home of Ritona town. Ritona is soon under attack by 3 more Gunmen but some quick (and insane) thinking by Kamina saves them..

In the aftermath of the previous battle, Kamina, Simon, and Yoko proceed to forage for provisions. However, a chance encounter with a formidable Beastman, Viral, and his powerful Gunman forces the Gurren and the Lagann to combine to form a new Gunman.

In order to take the fight to the Beastmen, Team Gurren sets out to strike a possible Gunmen rallying point. Along the way, they encounter the eccentric Black Siblings, as well as the prospects of having to defeat a multi-faced enemy.

Team Gurren's journey brings them to an underground village, where society mandates a humbling existence and the worship of Gunmen as divine beings. Team Gurren is joined by Rossiu and two children, Gimmy and Darry.

On their way to the Gunmen rallying point, Team Gurren decides to take a break at a hot springs getaway they find in the middle of nowhere, only to discover not everything is as it seems.

One of the Four Divine Generals of the Spiral King arrives to support Viral's Enki. Seizing the opportunity to land a great victory, Kamina and Simon pilot the Gurren-Lagann against seemingly overwhelming odds.

Having temporarily stalled Viral and Thymilph in the last skirmish, the Team Gurren and their newly arrived allies join forces as "Team Dai-Gurren" and launch a decisive strike against the regrouping Gunmen force.

Team Dai-Gurren managed to take control of the enemy fortress. After falling off a cliff, Simon notices a Gunman throw a capsule into a deep valley.

Nia learns of Kamina's significance in Simon's erratic behavior and offers him some stinging, yet honest advice, which draws the ire of an untrusting Yoko. With tension building within the Team, the crew scrambles to stand by Nia's side when Adiane returns to complete her stalled attack.

The Team falls into a trap set by the third member of the Four Divine Generals. They are taken captive and await execution underground.

On their way to Teppelin Team Dai-Gurren stumbles upon a vast ocean and must wait before crossing it. While the rest of the group play at the beach, Yoko starts to feel uneasy about Nia. After preparations are complete, Team Dai-Gurren begins their sea cruise, oblivious to the fact that an ambush is underway by Adiane and Viral.

Trying to do her part in the Team Nia decides to become a cook. Cytomander, the fourth member of the Divine Generals, launches an aerial attack on the Dai-Gurren. Simon leads the counter-attack on the Gurren-Lagann.

At last, Team Dai-Gurren arrives at the outskirts of Teppelin, but before aiming to the city, they must face a combined full-scale attack of Cytomander and Guame's forces.

The last of the Divine Generals falls and the city of Teppelin collapses, revealing a Gunman of gigantic proportions. Simon and Nia struggle to pass through it in order to confront her father, the Spiral King himself.

During the following seven years, a new civilization rises and prospers under Simon's leadership. However, these peaceful times come to end when the threat mentioned in Lordgenome's last words starts to reveal itself.

After the Anti-Spirals' first attack and Nia's astonishing announcement, Simon desperately searches for her while Rossiu finally completes his secret project.

Rossiu overthrows Simon and places him under arrest. But when the Anti-Spirals launch a large-scale attack on Kamina City, he is left with no choice but to let Simon fight the enemy using the Gurren-Lagann.

As Simon arrives in prison, he unexpectedly meets Viral, who comments on how far he has fallen. Meanwhile, the truth about Rossiu's emergency plan and the consequences for those left behind on Earth are revealed.

Yoko had been working on an island as a teacher until she learned of the situation at Teppelin and returned to break Simon free. Reunited, Simon and the members of Team Dai-Gurren launch themselves into space to protect the Arc-Gurren from the enemy forces.

The Gurren-Lagann and the Arc-Gurren join together to form the massive Arc-Gurren-Lagann and stop the Human Annihilation System.

The members of Team Dai-Gurren make their preparations for the imminent war against the Anti-Spirals, but Rossiu is missing. However, when Kinon tells Simon about what he really intends to do, both begin desperately searching for him.

Guided by Simon and Nia's feelings for each other, Team Dai-Gurren break through dimensions in search for the Anti-Spirals' homeworld. But after falling into the enemy's trap, they end up losing some of their most precious members, due to its vicious offensive.

Team Dai-Gurren tries in vain to escape the ocean of despair created by the anti-spirals. Only a sacrifice can save the others.

After merging with Lorgenome's ship and transforming into the gigantic Super Galaxy Gurren-Lagann, Simon and his friends successfully fight back against the enemy. However, the enemy traps them in a world of dreams and desires. Just when all hope seems lost, an ally from the past reappears.

Simon and Nia are reunited. Team Dai-Gurren uses their combined spiral power to merge everyone together and create the galaxy-sized Tengen Toppa Gurren-Lagann, engaging the Anti-Spirals in a decisive battle for the fate of the universe.
